PRODUCTION, PURIFICATION AND CHARACTERIZATION OF LACCASE FROM WHITE ROT FUNGUS.

:Laccases are oxidoreductases enzymes that belong to the sub class copper oxidases.They mainly produced by white rot fungi which involved in lignin metabolism. The objectiveof this research was effective utilization of agro wastes for economical production laccasefrom, Pleurotusflorida. Submerged fermentation employed enzymeand namely rice husk, wheat bran and sugarcane bagasse used as substrates. activityof enzyme analyzed spectrophotometrically at 420nm, using ABTS (2,2′-Azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) a substrate. maximum obtained with 9.9 IU/ml day 9. optimum temperature pH were found be 30˚C 7. best nitrogen source optimized ammonium sulphate enhanced up to11.57 IU/ml. large scale batch (1litre) already conditions 13460 IU/L ofenzyme. Purification performed molecular weight determined 90kDa through SDSPAGE. Characterization done terms pH, effect inducers inhibitors.
